(1) Difference and selection of flat bottom and round bottom (U-type and V-type) culture plates
Culture plates of different shapes have different uses. The flat bottom is usually selected for cell culture, which is convenient for microscopic observation, has a clear bottom area, and the height of cell culture liquid level is relatively consistent. Therefore, when doing MTT and other experiments, whether adherent or suspended cells, flat bottom plates are generally used. The flat bottom culture plate must be used to measure the light absorption value. Pay special attention to the material. The label "tissue culture (TC) treated" is used for feeder cells.
U-shaped or V-shaped plate is generally used when there are some special requirements. For example, in immunology, when two different lymphocytes are mixed cultured, they need to contact and stimulate each other. At this time, U-shaped plate is generally selected, because the cells will gather in a small range due to the action of gravity. The round bottom culture plate will also be used in the experiment of isotope incorporation. It is necessary to collect the culture of cells with a cell collector, such as "mixed lymphocyte culture". V-shaped plates are often used for cell killing, immunology and blood agglutination tests. The experiment of cell killing can also be replaced by U-shaped plate (after adding cells, centrifugation at low speed).
